Objectives: 1. To understand isolate and analyze the characteristics of ASC-EXO, and PSC-EXO
2. To evaluate the biological effects of ASC-EXO, and PSC-EXO on cell migration, proliferation, collagen synthesis, melanin inhibition, and anti-inflammatory activity in skin regenerative aesthetics.
Introduction: Adipose tissue-derived mesenchymal stem cell exosomes (ASC-EXO) and plant stem cell-derived exosome-like nanoparticles (PSC-EXO) have attracted significant interest for their therapeutic potential in regenerative medicine and aesthetics. This study explores the characteristics and biological functions of ASC-EXO and PSC-EXO, with an emphasis on their anti-inflammatory effects, enhanced cell proliferation, and promotion of collagen synthesis, highlighting their potential to improve skin health.
Materials / method: ASC-EXO and PSC-EXO isolated using the ExoSCRT tangential flow filtration (TFF) method. ASC-EXO were applied to in vitro assay models such as cellular uptake, cell proliferation and collagen synthesis in HDF, anti-inflammation assay in LPS-macrophage. In addition, the contents of ASC-EXO were analyzed through protein, miRNA, lipid profiling. PSC-EXO were applied to in vitro assay models such as cellular uptake, cell proliferation, and collagen synthesis in HDF in vitro model, anti-inflammation, and melanin synthesis. In addition, the contents of Plant-EXO were analyzed for miRNA and proteins.
Results: Characterization of ASC-EXO revealed a size range of 30-200 nanometers, with over 777 of the proteins identified by proteomic analysis. 132 of the miRNAs were identified by small RNA sequencing analysis. The top 20 miRNAs accounted for approximately 78% of the total, and the top five miRNAs let-7a-5p, let-7b-5p, miR-4301, and miR-23a-3p contributed to 46% of the counts. Mass spectrometry quantified 373 lipid species in ASC-EXO by lipidomics.
miRNA analysis revealed that Plant-EXO has about 30 kinds matched to human-specific miRNAs.
Conclusion: These findings suggest that both ASC-EXO and PSC-EXO have unique and beneficial properties for skin-related applications, highlighting their potential for development into therapeutic agents in regenerative medicine and aesthetic uses. The OMICS analysis of ASC-EXO and PSC-EXO provides a comprehensive perspective on enhancing skin quality. These data collectively show that Exosomes containing microRNA and proteins have multiple biological functions in skin-related assays such as fibroblast growth and melanin content in melanocytes.
